Morphometric Analysis of Mice Testicular Tubules after Administration of Malathion and Maca International Journal of Morphology
Bustos-Obregón,Eduardo; del Río,Fernando Costa; Sarabia,Luis.
Organophosphoric (OP) agropesticidas are amply used to increase food production. However, it has been verified that they induce alterations at testicular level related to the diminution of fertility in humans as in animals. On the other hand, different studies have been made to develop chemical or natural compounds that can induce an antagonistic effect to OP. In previous studies an extract from a plant, from the Peruvian Andes (Maca) has been recognized by its stimulating action on spermatogenesis. In the present study the effects of both external agents were evaluated on testicular sections of testis of adult male mice on a population of 52 mice CF1, divided at random in 4 groups (Control, Malathion, Maca, Malathion-Maca), with sacrifice intervals of...

Oral rapamycin attenuates atherosclerosis without affecting the arterial responsiveness of resistance vessels in apolipoprotein E-deficient mice BJMBR
Gadioli,A.L.N.; Nogueira,B.V.; Arruda,R.M.P.; Pereira,R.B.; Meyrelles,S.S.; Arruda,J.A.; Vasquez,E.C..
The objective of the present study was to assess the effects of the immunosuppressant rapamycin (Rapamune®, Sirolimus) on both resistance vessel responsiveness and atherosclerosis in apolipoprotein E-deficient 8-week-old male mice fed a normal rodent diet. Norepinephrine (NE)-induced vasoconstriction, acetylcholine (ACh)- and sodium nitroprusside (SNP)-induced vasorelaxation of isolated mesenteric bed, and atherosclerotic lesions were evaluated. After 12 weeks of orally administered rapamycin (5 mg·kg-1·day-1, N = 9) and compared with untreated (control, N = 9) animals, rapamycin treatment did not modify either NE-induced vasoconstriction (maximal response: 114 ± 4 vs 124 ± 10 mmHg, respectively) or ACh- (maximal response: 51 ± 8 vs 53 ± 5%, respectively)...

Oxidative stress and comet assay in tissues of mice administered glyphosate and ampa in drinking water for 14 days JBAG
Mañas,F; Peralta,L; Ugnia,L; Weyers,A; García Ovando,H; Gorla,N.
Excessive amounts of the herbicide glyphosate are incorporated daily to the soil and the ecosystems. AMPA is its major environmental breakdown product. In this study we determined the levels of thiobarbituric acid reactive substances (TBARs); quantified superoxide dismutase (SOD) and catalase (CAT) activity in liver, kidney, lung and heart, and performed the comet assay in blood and liver of mice administered glyphosate (40 or 400 mg/kg/day) or AMPA (100 mg/kg/day) in drinking water for 14 days. Exposure to glyphosate 400 mg/kg induced a statistically significant (p <0.05) decrease of SOD activity in heart and an increase in CAT activity in kidney. In the comet assay there were statistically significant differences in all the treatments and tissues...

Preliminary studies on the effects of orally-administered Transforming Growth Factor-beta on protozoan diseases in mice OAK
Namangala, Boniface; Inoue, Noboru; Sugimoto, Chihiro; 井上, 昇.
Transforming growth factor beta-1 (TGF-β1) is a pleiotropic cytokine with both pro- and antiinflammatory properties, depending on its environment and concentration. The present study evaluated the effects of orally-delivered TGF-β1 on mice parenterally-infected with various protozoan parasites. We report that while orally-administered TGF-β1 seems to confer partial protection against murine chronic babesiosis and acute trypanosomosis, no beneficial clinical effects were observed against acute babesiosis, malaria or toxoplasmosis. Taken together, these preliminary data suggest that the systemic effects conferred by exogenous TGF-β1 could be parasite speciesspecific. The variations in different parasitic infections could be due to (i) intrinsic differences...

Jalili,C; Khani,F; Salahshoor,M. R; Roshankhah,S.
Nicotine consumption can decrease fertility drive in males through inducing oxidative stress and DNA damage. The color of turmeric is because of a substance called curcumin for which some anti-oxidative and anti-inflammatory properties have been identified. In this study, various doses of curcumin (10, 30 and 60 mg/kg) and curcumin plus nicotine (10, 30 and 60 mg/kg) were administered intraperitoneally to male mice for 28 consequent days and reproductive parameters were determined. The results indicated that nicotine administration (0.5 mg/kg) significantly decreased testosterone level, count and motility of sperms, and testis weight compared to control group. However, increasing the dose of curcumin significantly increased reproductive indices in most of...

XU,PENGJUAN; LI,YOUYOU; YU,ZHICHAO; YANG,LIN; SHANG,RONG; YAN,ZIHANG.
Abstract: Triptolide, a purified diterpenoid from the herb Tripterygium wilfordii Hook.f., was widely used to treat many diseases. However, the hepatotoxicity of triptolide limited its clinical use. Research showed oxidative stress played an important role in triptolide-induced liver injury. To investigate the effect of vitamin C, which was one of the most effective antioxidants, on triptolide-induced hepatotoxicity and its potential mechanism in mice. In the present study, acute liver injury was induced by intraperitoneal injection of triptolide and vitamin C was orally administered. The results showed treatment with vitamin C prevented the triptolide-induced liver injury by reducing the levels of aspartate transaminase from 286.86 to 192.48 U/mL and...

Protective immunity of blood plasma fraction containing Babesia rodhaini exoantigens in mice OAK
Kamiya, Shiho; Takabatake, Noriyuki; Fujii, Kei; Okamura, Masashi; Iseki, Hiroshi; Yokoyama, Naoaki; Igarashi, Ikuo.
Babesia spp. are major hemoprotozoan parasites in animals. Exoantigens, which are soluble antigens released by the parasites during their asexual development, have been successfully applied for the development of a vaccine against babesiosis. In this study, we examined the ability of exoantigens to induce protective immunity in mice against a rodent Babesia parasite, Babesia rodhaini. The BALB/c mice immunized with the blood plasma fraction of B. rodhaini-infected mice (B. rodhaini exoantigens) showed a significant decrease in the lethal rate when challenged with B. rodhaini. Western blot analysis with anti-B. rodhaini exoantigen immune serum specifically detected at least eleven proteins in the lysate of B. rodhaini-infected erythrocytes. Next, a cDNA...

Role of berberine in ameliorating Schistosoma mansoni-induced hepatic injury in mice Biol. Res.
Dkhi,Mohamed A.
BACKGROUND: Schistosomiasis is caused by helminth parasites of the genus Schistosoma. Berberine chloride (BER), an isoquinoline alkaloid, has been used in vivo for its antiparasitic, antioxidant and hepatoprotective properties. In this study, the protective effect of BER and praziquantel has been compared for the extent of schistosomiasis-induced oxidative stress in hepatic tissue of mice. RESULTS: S. mansoni was able to induce inflammation and injury to the liver, evidenced (i) by an increase in inflammatory cellular infiltrations, dilated sinusoids and vacuolated hepatocytes, (ii) by decreased levels of alanine and aspartate aminotransferases and increased levels of alkaline phosphatase, γ-glutamyl transferase in the liver homogenate, (iii) by...

Sex-dependent differences in the activities of acetylsalicylic acid-esterases in mouse kidneys BJMBR
Benedito,M.A.C..
Acetylsalicylic acid (ASA), the most used drug worldwide, is hydrolyzed to salicylic acid and acetate by esterases present in tissues of several species including humans. Sex differences in drug metabolism by rodent liver are documented in the literature. In this paper we report a difference in the activities of the esterases (ASA-esterase I and II) in the kidneys of male and female mice. In this species there is no difference between males and females in liver ASA-esterases (ASA-esterase I: males 38.5 ± 7.9 (N = 5) and females 31.6 ± 7.6 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05; ASA-esterase II: males 77.3 ± 17.4 (N = 5) and females 61.4 ± 15.1 (N = 5) nmol of salicylic acid formed min-1 mg protein-1, P>0.05)....

Santos,Ed Wilson; Oliveira,Dalila Cunha; Hastreiter,Araceli; Silva,Graziela Batista; Beltran,Jackeline Soares de Oliveira; Rogero,Marcelo Macedo; Fock,Ricardo Ambrósio; Borelli,Primavera.
Obesity is a chronic inflammatory disease that affects millions of people worldwide. Most studies observe the effects of a high-fat diet (HFD) in 10-12 weeks. This work investigated the effects induced by a HFD administered for 6 weeks on the nutritional status of mice and some aspects of the inflammatory response in mouse peritoneal macrophages. Male Swiss Webster mice, 2-3 months of age, were fed a control diet or HFD for 6 weeks. After this period, the mice were euthanized, and peritoneal macrophages were collected for immunoassays and assessment of biochemical parameters. A HFD was associated with increased cholesterol, insulin resistance, C-reactive protein (CRP), leptin, and serum resistin levels. Lipopolysaccharide (LPS)- stimulated adipocyte...

Stimulatory factors for interleukin-12 production from murine splenic macrophages co-cultured with Babesia microti and Babesia rodhaini infected erythrocytes OAK
Hashiguchi-Kato, Rie; Thi, Pham Ngoc; Ohmori, Takashi; Tamahara, Satoshi; Katayama, Shinsuke; Shimada, Terumasa; Matsuki, Naoaki; Ono, Kenichiro.
Stimulatory factors for interleukin 12 (IL-12) production associated with Babesia microti and Babesia rodhaini infected erythrocytes were examined using an in vitro assay system established, since a remarkable increase of serum IL-12 concentration and differentiation of helper T cell (Th cell) into helper T cell type 1 (Th1 cell) was observed in early phase of infection with Babesia spp. in mice. To investigate direct stimulating activity of infected erythrocytes for IL-12 production, intact splenic macrophages were co-cultured with them and examined the expression of IL-12 mRNA by reverse transcription-polymerase chain reaction (RT-PCR). Both B. microti and B. rodhaini infected erythrocytes elicited IL-12 mRNA expression in co-cultured macrophages. Since...
